چکیده

These experiments were made to obtain more information about the surface of the red blood cell. Since "surface" means different things to different people, we shall define our meaning now. Strictly speaking, the surface studied by electrophoresis is the surface of shear between the cell (moving in the electric field) and the surrounding medium, for it is the potential at this surface which is the electrokinetic potential. But since this potential has its origin in the charged groups possessed by the cell membrane, we shall broadly use the term surface (unless we say otherwise) to mean that part of the membrane possessing these charged groups. That part of the membrane may be limited to the outermost constituent molecules of the membrane (i.e., to those molecules just inside the surface of shear), but we have no direct evidence of this. Also we have no data on the variations of charge density in different portions of the surface. The electrophoresis studies in this paper are divided into three parts. First, we have determined the mobility of human red cells as a function of the ionic strength at approximately constant pH. Secondly, we have determined the mobility as a function of pH at constant ionic strength for intact red cells, for the lipid of the red cell stroma, and for the protein of the stroma. Finally, we have determined the mobility of cells and ghosts under experimental conditions which cause changes in the mobility.

Electrophoretic studies of red cell extracts of stored blood.

It has been shown that red cells of blood collected in acid-citrate-dextrose (ACD) undergo changes in their dimensions, osmotic fragility, permeability and metabolism during storage at 4° C (1). Information concerning the effect of storage on the physical and chemical states of soluble red cell proteins, particularly hemoglobin, is not available. Such data may be of interest in view of the mark...

Electrophoretic Studies on Human Red Blood Cells

1. The electrophoretic mobility of unhemolyzed human red cells has been determined as a function of ionic strength at approximately constant pH in isotonic mixtures of glucose solution and saline-phosphate buffer solution. 2. Above an ionic strength of about 0.02 the cells behave as particles with a smooth surface of large radius of curvature. Below an ionic strength of about 0.02, changes of t...

The Electrophoretic Velocity of Human Red Cells, of Their Ghosts and Mechanically Produced Fragments, and of Certain Lipid Complexes

Ghosts prepared in CO(2)-saturated water from unwashed human red cells can be fragmented mechanically, but ghosts from thrice washed cells cannot. If the ghosts are prepared by freezing and thawing, this difference is not observed. The electrophoretic velocity varies also with the way in which the ghosts are prepared. The pH-mobility dependence of washed red cells flatten off to a plateau at pH...

Comparative Studies of High Contrast Fluorescence Imaging Efficiency of Silica-coated CdSe Quantum Dots with Green and Red Emission

Herein we report the possibility of using green and red emitting silica-coated cadmium selenide (CdSe) quantum dots (QDs) for remarkable stem and cancer cellular imaging, efficient cellular uptake and fluorescence imaging of semi and ultra-thin sections of tumor for in vivo tumor targeted imaging applications. The comparative studies of high contrast cellular imaging behaviours of the silica-co...

Agglutination by polylysine of young and old red blood cells.

The rate of agglutination by polylysine of young and old human erythrocytes was automatically recorded and correlated with measurements of the electrophoretic mobility of these cells. Old red cells have a reduced electrophoretic mobility as compared with young cells, as well as a higher rate of agglutination than young cells. Mild treatment of red cells with receptor destroying enzyme (RDE) whi...

Electrophoretic Mobility and Agglutinability of Red Blood Cells: a "new" Polymorphism in Mice

A quantitative method has been developed to determine agglutinability of mouse red blood cells. Tests with different inbred strains of mice revealed only two phenotypes. The same inbred strains were tested with the cytopherometer to determine the electrophoretic mobility of the corresponding red cells. Again, two phenotypes were uncovered, and faster mobility was found in the red cells that had...
